Adhesion of Human Mesenchymal Stem Cells and Differentiation of SH-SY5Y Cells on Amyloid Fibrils

چکیده
Amyloids are protein/peptide aggregates traditionally associated with many human disorders. However recent evidences suggest that amyloids are also capable of performing various native biological functions in host organisms. Many lower organisms including bacteria and algae use the highly ordered and adhesive property of amyloids for adhesion to host/substrates and/or for colonization, thus suggesting amyloids could serve as extracellular matrices (ECMs). Recently, we had reported that mammalian cells can adhere on amyloids using membrane-fibril interactions and integrin-based focal adhesion. In the current study, we demonstrate the nanotopography of amyloid fibrils for adhesion of human mesenchymal stem cells (hMSCs). Further, we also investigate the amyloid surface for differentiation of neuroblastoma cells SH-SY5Y. Our present study delineates the role of amyloid self-assembly in cell adhesion and differentiation.

A Comparative Study of Neuroprotective Effects of Human Adipose and Dental Pulp Derived Stem Cells on Hypoxic SH-SY5Y Cells Exposed to CoCl2
Background and purpose: Recent studies have demonstrated the promising effects of mesenchymal stem cells (MSCs) in some neurodegenerative diseases and proved their neuroprotective effects. But, the detailed pathways and the ability of MSCs from various sources has not been fully investigated. Materials and methods: Here, we isolated MSCs from two sources; adipose tissue and dental pulp, and co...

Objective(s) The use of co-culture cells as well as extra cellular matrix are among those strategies that have been employed to direct mesenchymal stem cell (MSC) bone differentiation in culture. In this regard, there is no study considering the effects of Matrigel on mesenchymal stem cell (MSC) in vitro bone differentiation. This was the subject of the present study. Materials and Methods ...
متن کاملThe Secretome of Bone Marrow and Wharton Jelly Derived Mesenchymal Stem Cells Induces Differentiation and Neurite Outgrowth in SH-SY5Y Cells
The goal of this study was to determine and compare the effects of the secretome of mesenchymal stem cells (MSCs) isolated from human bone-marrow (BMSCs) and the Wharton jelly surrounding the vein and arteries of the umbilical cord (human umbilical cord perivascular cells (HUCPVCs)) on the survival and differentiation of a human neuroblastoma cell line (SH-SY5Y). For this purpose, SH-SY5Y cells...
متن کاملIn situ fibrillizing amyloid-beta 1-42 induces neurite degeneration and apoptosis of differentiated SH-SY5Y cells
The progression of Alzheimer's disease is causatively linked to the accumulation of amyloid-β aggregates in the brain, however, it is not clear how the amyloid aggregates initiate the death of neuronal cells. The in vitro toxic effects of amyloid peptides are most commonly examined using the human neuroblastoma derived SH-SY5Y cell line and here we show that differentiated neuron-like SH-SY5Y c...
متن کاملA New Two Step Induction Protocol for Neural Differentiation of Human Umbilical Cord Blood-Derived Mesenchymal Stem Cells
Background: In this study, we examined a new two step induction protocol for improving the differentiation of human umbilical cord blood-derived mesenchymal stem cells into neural progenitor cells. Materials and Methods: Human umbilical cord blood-derived mesenchymal stem cells were first cultured in Dulbecco’s modified eagle medium supplemented with 10% fetal bovine serum in a humidified incu...
